Conditional Ablation of NKG2D Receptor in Mouse
NKG2D is an activating receptor expressed on NK cells, activated T cells and macrophages. It belongs to the C-type lectin-like family of receptors and interacts with H60, Rae1 and Mult1 ligands. The aim of this work is to inactivate mouse NKG2D receptor in a conditional and specific manner by the loxP/Cre recombination system in order to study its role in the development, activation, and homeostasis of the NKG2D expressing cell types in vivo. Therefore, we are developing a conditionally NKG2D mutated mouse strain by gene targeting. Taking in account that NKG2D is in some cases expressed only on subpopulations of cells (i.e. activated T cells and macrophages) we are also developing another mouse strain with the knock in of the EGFP gene into the NKG2D locus in order to trace the NKG2D expression. By generating a mouse having both alleles (NKG2D floxed and EGFP knock in/NKG2D knock out) together, we will be able to discriminate conditionally NKG2D deleted from non-deleted cells.
