Detection of human papillomavirus (HPV) in various lesions of oral cavity

Human papillomavirus (HPV) has been associated with oropharyngeal squamous cell carcinoma (SCC). It is a major risk factor of oral hyperplastic lesions development and further malignant transformation of some of these lesions. The purpose of this study was to examine the occurrence of HPV infection and the prevalence of different HPV genotypes in various oral lesions. During the period from 1995 to 2001, 146 oral specimens were collected by cytobrush from 47 men and 99 women (age range13-87 years) from 9 different topographic sites in the oral cavity. HPV DNA was detected by polymerase chain reaction (PCR) using two sets of universal primers (MY09/MY11 and L1C1/L1C2) and type-specific primers for HPV 6/11, 16, 18, 31 and 33 as previously described (Grce M et al., Anticancer Res 21: 579 ; 2001). Out of 146 specimens, 31 (20, 94%) contained one or more HPV types. The most frequently observed type was HPV 16 (35, 4%) that was predominantly associated with leukoplakia verrucosa, while HPV 6/11 and 31 were associated with oral focal hyperplasia and oral papillomas. Undetermined HPV DNA (types other than 6/11, 16, 18 and 31) was found in 13/31 (42%) cases and was equally distributed in various oral lesions (leukoplakia, erythroplakia, lichenoid reactions, Heck disease, condyloma acuminatum, smoker’ s palate). No preferential mucosal site for HPV DNA was observed. Our results strongly support the notion that HPV could be responsible for at least some oral hyperplastic lesion.
