Objectives: Although it is repeatedly demonstrated that invariant natural killer T (iNKT) cells perform an important function at the maternal-fetal interface, where during the pregnancy on villous and extravillous trophoblasts is expressed CD1d, the role of the hepatic NKT cells, during the pregnancy is still not well defined. Material and Methods: Since hepatic accumulation of these "primitive" lymphocytes might be induced by the syngeneic pregnancy, as well as by the treatment of non-pregnant mice by bacterial product -peptidoglycan-monomer (PGM) or PGM-Zn, in this study we tried to elucidate the effects of PGM-Zn in pregnancy. It was given both in vivo and in vitro, and changes were analyzed by flow-cytometric estimation of phenotypic and cytotoxic properties of hepatic and splenic mononuclear lymphatic cells (MNLC) against YAC-1, P815 cell lines and syngeneic thymocytes. Results: The data have shown that on the 16th day of syngeneic pregnancy in the liver accumulate CD3intermediate, NK1.1+ and IL-2Rb+ cells, which are markedly sensitive to PGM-Zn, since its in vivo application markedly increased the proportion of hepatic NKT cells, enhancing also the cytotoxicity of hepatic and splenic MNLC against the tested targets. Furthermore, the in vitro application of PGM-Zn additionally increased the cytotoxicity of effector cells obtained from pregnant mice, against NK-sensitive and LAK-sensitive targets, as well as against syngeneic thymocytes. Conclusions: The data emphasize that during the pregnancy in the liver are generated or accumulated forbidden clones of iNKT cells, which are sensitive to bacterial products, suggesting that their overexpresion in infection might contribute to unfavorable outcome of pregnancy.
