The influence of 4-hydroxy-2-nonenal on proliferation and differentiation of human osteosarcoma cells in vitro

Product of lipid peroxidation, 4-hydroxy-2-nonenal (HNE) is known to cause cell death when used in high concentrations, but in lower concentrations it can influence cell proliferation and differentiation. In our experiments we have used human osteosarcoma cells (HOS) to test influence of HNE on cell proliferation and differentiation. HNE caused dose-dependent inhibition of HOS cells proliferation measured by radioactive thymidine incorporation and increase in apoptotic cell death measured by TUNEL technique. When treated repeatedly with non-cytotoxic concentrations of HNE, decrease in spontaneous cell differentiation measured by formation of alkaline phosphatase positive nodules was observed. Also, differentiated cultures of HOS cells (cultures with alkaline phosphatase positive nodules) were more sensitive to inhibitory effect of HNE on cell proliferation than undifferentiated. Those two cell cultures differed also in GSH content, which can explain different sensitivity to HNE. Data obtained indicated that HNE could prevent HOS cells differentiation, probably by reducing number of differentiated HOS cells in cell cultures. Also, process of HOS cells differentiation caused subtle changes in cell metabolism resulting in decreased GSH content in those cells.
