Sinergistički učinak čimbenika rasta FGF-a i NGF-a na regulaciju rasta kultiviranih zametaka sisavaca
FGF plays an important role in vivo in cell proliferation and differentiation in embryogenesis, tissue regeneration, CNS development, wound healing and tumor progression. NGF is significant for the survival and development of symphatetic and neural crest-derived sensory neurons. The growth of rat embryo in vitro in response to FGF and NGF signals and their possible synergistic action was investigated. Rat embryos in gastrulation phase were treated with 100 ng/ml and 200 ng/ml of FGF and NGF, and with both agents combined in serum-free (MEM)and serum supplemented medium. Growth of explants was evaluated by measuring average size of explants during 14 days of culture. Both concentrations of NGF impaired growth, while FGF stimulated it, even it was not always significantly different from control groups in a paticular measure-day. When NGF and FGF were applied together in serum-supplemented medium, the growth was significantly improved compared to control and other experimental groups. It seems that NGF and FGF may act synergistically, thus improving growth of embryos in serum-supplemented medium.
