Decidual NK cells use Fas/FasL cytolytic pathway
Decidual lymphocytes (DL) infiltrate human first trimester pregnancy decidua containing mainly decidual NK (dNK) cells (CD56bright+). dNK are activated and express high number of perforin in their granules. The aim of our study was to analyse molecular mechanisms involved in short term cytolytic activity of dNK cells. Decidual tissue was obtained from elective termination of normal human 6-10 week old pregnancies. Decidual mononuclear cells (DMC) were isolated by enzymatic digestion and density gradient centrifugation. DL were collected from the suspension of DMC immediately or following 18 hours culture with or without Th1 cytokines. The cytolytic activity of DL against K562, P815 and P815Fas cells was analysed by PKH-26 (red) cell linker kit and flow cytometry. dNK cells were isolated by positive magnetic cell separation method. Direct and indirect immunofluorescency was used for surface antigen labelling. RESULTS: Non-stimulated DL efficiently lyse NK sensitive K562 cells and NK resistant P815Fas and P815 cells. Th1 cytokines are not able to up-regulate significantly the cytolytic activity of DL against P815 and P815Fas cells. However, non-stimulated DL lyse significantly more P815Fas than P815 cells. Concanamycin A and anti-FasL antibody significantly down-regulate cytolytic activity of cultured dNK cells against P815 targets. CONCLUSIONS: First trimester pregnancy DL in vitro efficiently use both perforin and FasL mediated cytotoxicity pathways. This indicates that in vivo dNK cells, if activated could use perforin and FasL cytotoxicity against intracellularly parasitized and transformed cells or trophoblast cells.
