Olive and corn-oil enriched diets as modulators of tissue mineral content in the submandibular gland of partially hepatectomized mice

Fast liver growth, which follows after partial hepatectomy (pHx), is well-defined process, which involves the concerted action of extra and intracellular factors resulting in cell replication and its inhibition at the time when the entire liver mass is restored. Owing to our previous findings that essential trace elements influence liver growth and immune functions, as well as that particularly the submandibular gland (SMG) produces significant quantities of growth factors involved in reparatory processes (EGF, NGF, TGF-beta, kallikreins, insulin-like growth factor family), in this study we estimated: 1) the tissue mineral dynamics in SMG after pHx and 2) the influence of olive and corn-oil enriched diets, containing different quantities of mono- and polyunsaturated fatty acids (MUFA and PUFA) on these events. For these purpose male BALB/c mice, aged 2-4 months, were during the 3 weeks fed by diets enriched with olive or corn oil (5% addition to standard pellet, w/w) and then subjected to 1/3 pHx. One, 2, 7 and 15 days later mice were sacrificed, and tissue content of Zn, Fe, Mg and Ca was determined in the SMG, and in the remaining liver, using the microwave digestion and ICP spectrometry. The data showed that in early phase of regeneration in the liver accumulated Zn and Fe, while in the SMG accumulated Zn and particularly Ca, and decreased the level of Mg. Moreover, in SMG a high correlation was found between the tissue concentrations of Zn and Ca on 7th p.o. day (r=0.82), Fe and Ca on 15th p.o. day (r=0.98), as well as between the Mg and Ca on the 1st and 15th day after pHx (r=0.99). In the later phase of regeneration a highly significant negative correlation was found between the level of Zn, Fe, Mg and Ca and intensity of liver growth after pHx (r=-0.99). The diets enriched by olive oil, with oleic acid (C18:1, n-9), and corn oil, containing mostly linoleic acid (C18:2, n-6) significantly decreased the tissue levels of Zn, Fe and Ca in SMG, increasing the regenerating index on 7th p.o. day. Furthermore, in mice fed by corn-oil diet, a positive correlation was found between the Mg concentration in the SMG and remnant liver mass on the 15th day of regeneration. The data imply that trace elements regulate the activities of SMG, and that diets containing MUFA or PUFA affect these dynamics, regulating the degree of oxidative stress and lipid peroxidation, activities of matrix metalloproteinases, signal transduction pathways, cytokine production etc.
