Breakdown of Chlorophyll: Biochemical Properties of Red Chlorophyll Catabolite Reductase

Red chlorophyll catabolite reductase (RCCR) is responsible for the biosynthesis of fluorescent chlorophyll catabolites (FCCs) by reduction of the C20/C1 double bond in the red chlorophyll catabolite (RCC), a putative intermediary catabolite in chlorophyll breakdown. The mechanism of reduction catalyzed by RCCR and means of regio and stereo control are still unknown for this class of enzymes. Cloned RCCR (Brassica napus) used in this study was expressed in E. coli, isolated and purified by the attached His-tag. The binding of RCC to the enzyme was studied by UV/Vis and circular dichroism spectroscopy in the absence of a reducing agent. RCCR belongs to the class of reductases that use reduced ferredoxin but appear not to need redox-active cofactors. Electro-synthetic studies were undertaken to determine whether RCC and its derivatives have an inherent propensity for one-electron reduction and protonation to give FCCs.
