Before the beginning of the Human Micronucleus Project in almost no attention was paid to the occurrence of nuclear buds in cytokinesis-blocked cells. However, evidence has been accumulating that nuclear buds arise from the elimination of amplified DNA and possibly elimination of DNA-repair complexes. Since than only few results of micronuclei assay that included scoring of the buds were published. In the present study micronucleus assay have been used to assess possible the chromosome-damaging activity of the pesticide formulation Deherban A® ; ; containing 500 mg/ml of 2, 4-dichlorophenoxyacetic acid as an active ingredient. Human peripheral blood lymphocytes were treated with the pesticide for 24 hours in the absence and in the presence of the rat liver microsomes as metabolic activation system. Two different concentrations of pesticide expressed as the concentration of the active ingedient 2, 4-D have been tested (0, 4 mg/ml and 4 mg/ml). In addition to micronucleus assay, the frequency of nuclear buds in binucleated lymphocytes was analyzed. The total number of micronuclei and nuclear buds was significantly increased in the treated lymphocytes compared to the control for both concentrations tested. The number of binucleated lymphocytes with more than 1 micronucleus was higher in treated lymphocytes compared to the control but that difference was not statistically significant. The presence of the metabolic activator caused a significant increase in the number of micronuclei compared to the treatment in the absence of the S9 mix for both concentrations tested. The number of nuclear buds was significantly elevated by the presence of the metabolic activator only at the higher concentration of Deherban A® ; ; . This results sugest that in risk assessment studies in vitro, which should simulate the exposure conditions, entire pesticide formulations instead of pure active ingredients should be used, as it was the case in the paper presented.
