TLC Identification of Metal Components in Base Alloys
Objectives: Knowledge of the composition of incorporated alloys is the best way to avoid polymetallism, allergic reactions and galvanism with prosthetic restoration. We are reporting an analytical procedure for detection of components in dental alloys based on cobalt and nickel. Methods: The method involves microanodic sampling in situ, separation and identification of components by means of thin-layer chromatography on pre-coated cellulose layers. The sampling was performed with the HCl: H2 O=1: 10 (v/v) electrolyte. The optimum solvent system was: iso-AA:HCl(conc.):ACN=53.6:2.7:43.7 (v/v), and for visualization: quercetin (0.1%) propanolic solution + dimethylglyoxime ethanolic solution (1%) + NH3 vapors. Results: A very good separation and detection of Ni, Cr, Co, Mo and Be based on different Rf -values and colours of spots was achieved. Conclusion: Described method of sampling, microanodic dissolution and identification is fast non-destructive, and provides detection of suspected metals in incorporated permanent prosthetic restorations. Standardization of in situ anodic sampling procedure will enable accurate quantitative determination using video densitometry
