DNA integrity determination in marine mussel Mytilus galloprovincialis by Fast Micromethod&reg ; - a long-term field study

This study was focused toward the adaptation of the previously developed Fast Micromethod&reg ; for DNA damage determination in marine invertebrates for the establishment of biomonitoring assessment. The Fast Micromethod&reg ; detects DNA damage (strand breaks, alkali-labile sites, cross-links and incomplete excision repair) and determines DNA integrity in mussel Mytilus galloprovincialis Lam. gills homogenates in single microplates. In vivo experiments with mussel Mytilus galloprovincialis gills homogenate by some xenobiotics presented herein indicated the decrease of the DNA integrity. The DNA cross-links were detected by using their known chemical inducers. In vitro action of bleomycin (bleomycin-Fe(II) complex) with mussel gills homogenate were used for induction of DNA strand brakes and yielded high negative strand scission factor (SSF) values. Present approaches to environmental pollution biomonitoring programs are focusing on investigations at regional and local levels in an attempt to precisely define the nature and extent of any potential environmental crisis. The Fast Micromethod&reg ; was used in the Adriatic Sea monitoring program to detect the effect of mixed marine pollutants on DNA integrity in the mussel Mytilus galloprovincialis gill tissue. The mussels were collected along the eastern Adriatic coast over a long-term period. The strand scission factor (SSF) values, as a measure of DNA integrity, have shown that the Split region (Kaštela bay) is the most contaminated area by genotoxic agents. The estimation of directional changes of harmful effects in the ecosystem is expected to be significant in the context of genetic and biomonitoring not only on local levels but also on a global scale.
