Influence of selenium on DMSA efficiency in mercury chelation and elimination in rats
Simultaneous administration of mercury and selenium causes redistribution of mercury in the organism and decreases mercury excretion. The purpose of this work was to evaluate the efficiency of the chelating agent meso-2, 3-dimercaptosuccinic acid (DMSA) in mercury removal from the body of rats when mercury is co-administrated with selenium. Female Wistar rats, 8 weeks old, were randomly divided into four groups which were assigned: 1. Hg ; 2. Hg + DMSA ; 3. Hg + Se ; 4. Hg + Se + DMSA. The animals were given a single intraperitoneal injection of mercuric chloride or equimolar mixture of mercuric chloride and sodium selenite (1.5 microM/kg body weight). DMSA (500 microM/kg) was given orally, 30 minutes after the administration of mercury and selenium. The animals were euthanized 24 hours after the treatment and the concentrations of mercury in the kidney, liver, and 24-hour urine were determined using cold vapour atomic absorption spectrometry (CV-AAS). The simultaneous administration of mercury and selenium led to a change of mercury distribution in the body, by lowering mercury accumulation in the kidney and increasing mercury accumulation in the liver. Selenium also caused a decrease in the level of urinary mercury excretion. DMSA efficiently increased excretion of mercury into urine, but when animals were simultaneously treated with mercury and selenium the rise of mercury excreted in the urine due to the DMSA treatment (comparison group 4. vs. group 3.) was lower than in animals where mercury was given without selenium (comparison group 2 vs. group 1). It is concluded that selenium decreases the efficiency of DMSA in mercury removal from the body of rats.
