Yeast non-replicative plasmids and strain construction
Stable transformation with yeast non-replicative vectors relies on homologous recombination between yeast chromosome and plasmid molecule. We show that UV-irradiation of double-stranded plasmids stimulates this process. Irradiation of either double or single-stranded non-replicative plasmids does not lead to detectable mutagenesis. Transformation by conversion of point mutations is favored if single-stranded plasmids are UV-irradiated. Linearized single-stranded plasmids may even convert the insertions of the Ty element (6.1 kb).
