POLYFLAMIN - CHARACTERISATION AND IMMUNOSTIMULATING PROPERTIES

Polyflamin is the mixture of disintegrated bacteria (Escherichia coli, Staphylococcus aureus, Streptococcus hemolyticus). It was produced at the Institute of Immunology up to 1993 and was used for the treatment of inflammations of uterus and ovaries as a non-specific immunostimulator. Its immunostimulating properties have not been sufficiently documented and studied. Many similar bacterial preparations, available and registered for years in Europe and Japan, have been widely used for diverse indications. It was, therefore, plausible to attempt the further characterisation of polyflamin prepared at the Institute of Immunology and to study its immunostimulating properties. Polyflamin is produced as a suspension quantified according to the number of bacterial cells. Each batch was examined by thin layer chromatography and HPLC and protein content was determined. Distribution of molecules according to molecular mass was studied by gel chromatography. Immunostimulating properties were studied in vivo. Mice were immunised with ovalbumin (OVA) and polyflamin, control group only with OVA and the group immunised with OVA and peptidoglycan monomer (PGM) was a positive control. Specific anti-OVA IgG was determined in sera of immunised mice using appropriate enzyme immunoassay (ELISA) developed and standardised in the laboratory. We have demonstrated that polyflamin stimulates the production of anti-OVA IgG in mice in two separate experiments. The effect of polyflamin was more pronounced than the effect of PGM. Further experiments in different models seem to be justified.
