Dynamic organization of the actin cytoskeleton in Dictyostelium
An overview of recent studies on function of actin-binding proteins conducted in amoeboid cells of Dictyostelium discoideum will be given. Creation of mutants and their phenotypic analysis have led to significant advances in our understanding of how actin-binding proteins function in a cellular environment. Monitoring of the dynamic localization of GFP-fusion proteins has provided important clues about the turnover rates and transport processes within the actin cytoskeleton, in particular by observation of spontaneously occuring spatio-temporal patterns and by FRAP experiments. Cryoelectron tomography of intact eukaryotic cells is an emerging technique that enables examination of the native cytoskeleton structure at an unprecedented detail. Pioneering results obtained from Dictyostelium cells will be presented and prospects of the method discussed.
