Determination of the coding and noncoding nucleotide sequences of original Edmonston-Zagreb master seed and current working seed
Edmonston-Zagreb vaccine strain is derived from a single clinical isolate of measles virus called the Edmonston strain and used for production of measles vaccine. Protein coding and noncoding sequences of the original Edmonston-Zagreb master seed, manufactured in 1967, and working seed D_24 2/99 lot, manufactured in 1999, were determined directly from DNA fragments generated by RT-PCR and sequenced by automated sequence apparatus. The coding and noncoding sequences of the Edmonston-Zagreb master seed and the working seed D_24 2/99 do not differ. The sequences are compared to reference strains, i.e. Edmonston wild type AF266288 and Edmonston Zagreb AF266290 (Parks et al., J. Virol. 75: 910-920, 2001). Differences were found in at least 29 and 6 nucleotide positions respectively. We suppose that such a substantial number of observed differences could be due to additional four passages of Edmonston-Zagreb AF266290 in Vero cell culture (Parks et al., J. Virol. 75: 910-920, 2001).
