Role of retinoic acid and DNA methylation in differentiation of mammalian embryo in vitro.
To explore a possible relationship between DNA methylation and induction of differentiation by retinoic acid during embryonic development we applied 5-azacitidine and RA to rat embryos cultivated in vitro. Cultivation of gastrulating rat embryos without extraembryonic membranes was performed at the air-liquid interface for two weeks. When 5azaC and RA were applied, differentiation in cultivated embryos changed. Differentiation of the cartilage and epidermis was inhibited but differentiation of the glandular epithelium and mucous metaplasia was enhanced.
