Detection of HPV from genital sites in asymptomatic men by RFLP and Digene Hybrid Capture II
Genital human papillomavirus (HPV)infection is one of the most common sexually transmitted diseases in both women and men. In men HPV infection can be subclinical or HPV can induce various clinical manifestations. Diagnostic approach to subclinical HPV genital infections in men is not standardised and importance of HPV detection in asymptomatic men is still controversial. The aim of our study was to compare detection of genital HPV in asymptomatic men by Digene Hybrid Capture II and RFLP. Patients: 93 asymptomatic men whose female partners had various cytological changes (CIN I-III). Samples were taken from predilectic genital sites:preputial cavity, glans, sulcus coronarius and urethra. All samples were taken with Digene hybrid Capture II collection kit. Digene Hybrid Capture II and RFLP were done from the same sample. Methods: all samples were analysed according to manufacturers instructions for Digene Hybrid Capture II. RFLP was done with A1uI restriction endonuclease after amplification with primer MY 09 and MY 11 according to Contorni an coauthors. Results:Out of 93 samples taken from asymptomatic men 80 were negative by both methods. 13 were HPV positive by RFLP, of them 8 were low risk positive, 4 were high risk positive and 1 had indeterminative electrophoretic pattern. Out of 13 HPV positive by Digene Hybrid Capture II, 6 were low risk positive, 3 were high and low risk positive and 4 were high risk positive. Discrepant results were for 2 low risk positive and 1 indeterminative sample by RFLP, all these samples were low and high risk HPV positive by Digene. Conclusion: there is accumulating evidence taht asymptomatic men are important reservoires of HPV. Molecular detection methods are primarely adapted for screening women. Further studies are needed to evaluate HPV detection by Digene HYbrid Capture II and RFLP in asymptomatic men
