Proportion of lymphoid and myeloid cell subsets in mesenteric lymph node of pigs immunized with F4ac^+ nonenetrotoxigenic Escherichia coli strain
To evaluate the immunogenicity of F4ac^+ non-enterotoxigenic Escherichia coli (non-ETEC) vaccine candidate strain in 4-week-old pigs we have used monoclonal antibodies (mAbs) reactive with swine leukocyte CD/SWC antigens to determine eventual changes in the immunophenotypic expression of these molecules and the proportion of CD+/SWC+ immune cells from mesenteric lymph node (MLN) by flow cytometry. Intragastric immunization of pigs with 10^10 CFU/ml of F4ac^+ non-ETEC strain Abbotstown A induced changes in the proportion of lymphoid and myeloid cells in MLN of immunized pigs as compared to those recorded in unimmunized pigs. The presence of antigen-specific T (CD3, CD4, CD8, CD25, CD45RC, CD45RA) and B cell (CD21, CD45RC, CD45RA) subsets and antigen presenting cells (APCs ; SWC3, CD14) demonstrates that the compartment of gut-associated lymphoid tissue of weaned pigs, i.e. MLN, is functionally organized for the expression of immune responses to the bacterial immunogens, such as F4 fimbriated non-ETEC tested as a potential source vaccinal antigen.
