Pentadecapeptide BPC 157 in stroke rats counteracts stress gastric lesions, thrombosis and intracranial, portal and caval hypertension, and aortal hypotension
Aim. Recently, after bilateral clamping of the common carotid arteries, the subsequent application of the BPC 157 in reperfusion documented the resolution of the neuronal damages sustained in the brain, resolution of the damages reflected in memory, locomotion, and coordination disturbances, with the presentation of the particular genes expression in hippocampal tissues (Brain Behav. 2020 Aug ; 10(8):e01726). Likewise, BPC 157 administration, by rapid activation of the collateral vessel pathways, rapidly reversed major venous occlusion syndromes, inferior caval vein syndrome, Pringle maneuver, ischemia, reperfusion, and Budd-Chiari syndrome (Vascul Pharmacol 2018, World J Hepatol 2020, World J Gastrointest Pathophysiol 2020). Thereby, this beneficial effect may resolve the adverse effect of temporary bilateral clamping of the common carotid arteries, ischemia, reperfusion, and counteract the consequent intracranial (superior sagittal sinus), portal and caval hypertension, and aortal hypertension, organ lesions, in particular stress hemorrhagic gastric lesions, arterial and venous thrombosis, which may appear in ischemia and reperfusion. Methods. Using described bilateral clamping of the common carotid arteries procedure (Brain Behav. 2020 Aug ; 10(8):e01726), ischemia (I) (20 min, medication (/kg, ig) BPC 157 (10μg, 10ng) or saline (5ml) ig at 1 min ischemia-time), and reperfusion (R) (20 min, medication (/kg, ig) BPC 157 (10μg, 10ng) or saline (5ml) at 1 min reperfusion-time) were performed. Results. Without therapy, the severe blood pressure disturbances occurred immediately, and were permanent. For instance, at 20 min ischemia- time and at 20 min reperfusion-time, controls presented consistent intracranial hypertension (17±2 (I) - 20±2 (R) mmHg in superior sagittal sinus), portal (32±4 (I) - 33±4 (R) mmHg) and caval (25±4 mmHg (I) - 22±4 (R)) hypertension and aortal hypotension (80±3 - 82±4 (R) mmHg). Grossly, we notedseverely congested stomach and major hemorrhagic lesions (sum of the longest diameters, 16±4 (I) - 20±4 (R) mm), and at 20 min reperfusion-time, clot formation (g) in the abdominal aorta (0.0084±0.0009), superior mesenteric artery (0.0052±0.0008), superior sagittal sinus (0.0036±0.0005), inferior caval vein (0.0201±0.0005), portal vein (0.0101±0.0008), superior mesenteric vein (0.008±0.0009) and lienal vein (0.0014±0.0007), comparable to these in the ischemia period. BPC 157 administration counteracted intracranial hypertension (i.e., 10ng) (-6±2 (I) - -10±2 (R) mmHg in superior sagittal sinus), portal (8±4 (I) - 7±4 (R) mmHg) and caval (7±2 mmHg (I) - 5±2 (R)) hypertension and aortal hypotension (98±5 - 101±5 (R) mmHg). Likewise, BPC 157-rats had no stomach lesions, and venous thrombosis and arterial thrombosis were almost annihilated. Conclusion. Pentadecapeptide BPC 157 should be further tested in clinic.
