Study of host-guest complexes of cyclodextrins and ferrocene uracil derivatives

Cyclodextrins (CDs) are cyclic oligosaccharides which provide an interesting organic host system, since they have a hydrophobic inner cavity available to form non-covalent host-guest inclusion complexes with a wide variety of organic molecules of appropriate shape and size. It is known that cyclodextrins show abillity to improve drug water solubility by binding the hidrophobic moiety of the molecule inside of its cavity, such as ferrocene.1 Ferrocene shows different binding orientations as well as binding constants for alpha, beta and gama cyclodextrine in water.2 Environment of ferrocene can influence their binding to cyclodextrines.3 Recent research shows that ferrocene uracil derivatives have shown great biological activity.4 However, their high lipophilicity limits the application. Therefore, complex formation with β-cyclodextrin, γ- cyclodextrin, modified (2-hydroxypropyl)-β- cyclodextrin and suggamadex with various ferocene uracyl derivatives with different R groups in water were studied (figure 1). Complex formation and interactions were observed by UV-Vis spectroscopy. Binding constants were calculated based on titrations with outcome in range between 2-3.
