Deep eutectic solvents and non-conventional technologies for blueberry peels extraction: kinetics, anthocyanins stability and antiproliferative activity

In recent years, food industry by-product’s high added value compounds recovery using non- conventional technologies is gaining increasing attention, since it can reduce environmental impact and ensure economic benefits.1 In the light of the aforesaid, two emerging extraction methods for anthocyanins recovery from blueberry processing residues were studied to obtain a novel antitumor agent. Firstly, five natural deep eutectic solvents (NADES) have been tested for anthocyanin extraction and the eutectic systems shelf-life was monitored over 9 months. The most suitable NADES was implemented in microwave- (MAE) and ultrasound-assisted extractions (UAE). Extraction kinetics was described using Peleg’s model. Both technologies enabled the performance and gave a result superior to that of conventional extraction performed using hydroalcoholic solution. MAE and UAE yielded up to 25.83 and 21.18 mg/gmatrix of total anthocyanin content, respectively, after 15 and 30 min. Finally, extracts antiproliferative activity was determined by an MTS assay on human tumour HeLa cells against human skin cells. Non-conventional extracts exhibited strong antiproliferative activity, much greater than that of their conventional analogue. Cell-death type was evaluated by flow cytometry, and apoptosis was found to be the primary cause of tumour cell death.
