Investigation of antiproliferative activity of copper(II) complex with chromone-2-carboxylic acid “in vitro”
Copper (II) complex with chromone-2-carboxylic acid (ligand = chromocarb) was synthesized by the methodof classical solution chemistry. The ligand belongs to the group of hydroxypyrones, which are known for thefollowing properties: low toxicity profile, easy availability and high functionality. They exert high affinity toward divalent and trivalent metal cations (Cu2+, Fe2+, Fe3+) forming potential therapeutic and diagnostic metallopharmaceuticals (especially cytostatics). Antiproliferative activity of ligand and copper (II) complex was evaluated by the MTT (3-(4, 5- dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide) assay on 7 cell lines obtained from the American Type Culture Collection (ATCC). The malignant cell lines were: Caco-2 and HT-29 (colorectal adenocarcinoma), MDA-MB 231 (human breast adenocarcinoma), KATO III (human gastric adenocarcinoma), Hep-G2 (hepatocellular carcinoma), NCI-H358 (bronchioalveolar carcinoma). The normal cell line was MRC-5 (human fibroblasts, from lung). The complex was dissolved in DMSO at a concentration of 10-2 M and applied on cells at final concentrations as follows: 10-5 M, 10-6 M, 10-7 M. All experiments were performed in triplicates. Evaluated copper (II) complex suppress cell growth of Hep-G2 cells at highest concentration (10-5 M) by 64, 8 % followed by NCI- H358, MRC-5 and HT-29 (45, 6% ; 45, 3% ; 43, 4%). Inhibitory effect of copper (II) complex on KATO III, MDA-MB 231 and Caco-2 is less than 25 % indicating low anti proliferative efficacy. Opposite to complex, the ligand itself does not exert inhibitory activity toward cell proliferation of investigated cell lines. According tothe observed results it can be concluded that the synthesis of the complex was successful and that the new compound demonstrates potential antiproliferative active.
