The discovery of genetic alterations in oncogenes and tumor-suppressor genes, which accompany tumor formation in a wide variety of human tumor types, has encouraged the search for genes that may promote or suppress tumor spread and metastasis. Among these, nm23 seemed to be the most promising candidate for a gene with metastasis-suppressing function. The purpose of this study was to evaluate whether the expression of the nm23-H1 protein or loss of heterozygosity (LOH) of the nm23-H1 gene is associated with colon cancer tumor progression. Praffin tissue sections were analyzed immunohistochemically. DNAs isolated from normal and tumor tissue were used for LOH analysis. To analyze LOH at the nm23-H1 gene we used VNTR marker located in untranslated 5' region of the nm23-H1 gene. Of 102 adenocarcinomas that were examined, 58.8% showed a weak positive staining for nm23-H1 protein. Negative correlation was found between nm23-H1 positivity and histological grade of tumors. In VNTR analysis 70.2% of patients were infomative and 27.4% of tumors demonstrated LOH at nm23-H1 locus. Positive correlation was found between the nm23-H1 LOH and histological grade of tumor as well as with Dukes' stage of tumor samples. The findings of this study suggest that more frequent nm23-H1 protein expression in early stages plays an important role in suppressing metastasis in sporadic colon cancer while at later stage both, less frequent nm23-H1 protein expression and LOH of the nm23-H1 gene play important role in colon cancer progression and metastasing.
