Nasturtiium officinale R. Br. in vitro cultures as a potential source of biologically active secondary metabolites and bioelements

The object of extensive biotechnological, phytochemical and biological works was a partially protected (acc. IUCN) species of the Brassicaceae family – Nasturtium officinale R. Br. (watercress). The studies concerned the optimization of conducting various types of in vitro microshoot cultures of N. officinale (agar, agitated and bioreactor), in terms of testing the effect of PGRs, cultivation period, light conditions, elicitation and media supplementation with biosynthetic precursors in order to obtain high production of metabolites (e.g. glucosinolates, phenolic acids and flavonoids) and bioaccumulation of elements. As part of the work, biological activity (antioxidant, anti-inflammatory, antimicrobial, cytotoxic and tyrosinase inhibition) of extracts from experimental cultures was assessed. High production of 4-methoxyglucobrassicin (max. 399 mg/L) and gluconasturtiin (max. 98 mg/L) was obtained in extracts from cultures grown in Plantform bioreactors. The extracts showed a significant inhibition of COX-2 (75.40%) and a strong antibacterial effect (especially vs. Helicobacter pylori) and a considerable antioxidant effect. The in vitro cultures showed a high bioaccumulation capacity for element ions (especially magnesium and calcium). The obtaining results of the research indicate the possibility of using in vitro cultures of N. officinale as a potential source of health-promoting substances, an alternative to the material obtained from in vivo conditions.
