Resistance of mouse myeloid leukemia cells to apoptosis: involving of c-Myc, Bcl-2 and p53

Background and Purpose: Before, we found an unusual resistance of mouse myeloid leukemia (ML) cells to high dose (1.280 J/m2) of UVC light. In the present work we investigated if resistant fraction of ML cells could die by apoptosis. Moreover, we investigated the possible role of oncoproteins (c-Myc, Bcl-2) and tumor suppressor protein (p53) in UVC-resistant phenotype. Materials and Methods: RFM/Rij Zgr mice, 3-4 months old, were used. After injection with 106 of living leukemia cells, the disease came to terminal phase. Spleen cells of leukemic mice and healthy mice (control) were exposure to UVC (dose from 5-1.280 J/m2), and 4 or 24 hours later is determined the number of viable and apoptotic cells. The presence of c-Myc, Bcl-2 and p53 proteins was detected by immunocytochemical method. Statistical analysis was performed using Model 1 ANOVA, followed by a Bonferroni test. Results: Some fraction of ML cells and spleen cells of healthy donors survived 1.280 J/m2. Mostly of leukemia cells did not enter in apoptotic way of death, what is different to healthy spleen cells. In that case we observed a live “ monster” cells, without apoptotic feature. In terminal phase of leukemia, the percentage of positive cells to all examined proteins are a lower than in healthy donors. UVC had not significant influence on the percentage of positive cells. Conclusion: Some fraction of ML cells can survive used doses of UVC. Destroyed levels of investigated proteins contribute to transforming phenotype and resistance of ML to UVC and apoptosis.
