Response of free lung cell populations from rats challenged by microwave irradiation

The aim of this study was to determine the number and type of free lung cells caused by whole body irradiation of rats with 2.45 GHz cw (continuous wave) radiofrequency microwave. Twenty of forty adult male Wistar rats were divided into four subgroups that received 2, 8, 13 and 22 irradiation treatments of two hours each. During the treatment, animals were kept in Plexiglas cages. The microwave irradiation frequency was 2.45 GHz with a power density of 5-15 mW/cm. The microwave source was a modified Micro-Chef Moulinex generator. The rectal body temperature was measured before and after each exposure treatment. Sham-exposed rats were used as controls. Control animals were also divided into four subgroups to be sacrificed on indicated days. Animals were euthanized by exsanguinations under pentobarbital anesthesia, on experimental days 1, 8, 16 and 30. Free lung cells were obtained by bronchoalveolar lavage of each lung. Cell response was determined by standard laboratory methods. The absolute number of retrieved cells was determined by a hemocytometer. The share of alveolar macrophages and their morphological features were determined by light microscope under oil immersion (magnification 1:1000). In comparison to the non-treated animal group, the total cell count and the share of alveolar macrophages decreased in the treated animal group during the course of the experiment. The significant decrease of total cell number and macrophage count was time- and dose-dependent, as was the increase of polynucleated cells. Findings point out that the observed free lung cell response might be a sensitive bioindicator of microwave irradiation
