Morphological changes in adipocytes of pregnant rats on high fat - high sugar diet

Aim. To investigate the effects of high fat high sugar diet (HFHSD) on morphological changes in subcutaneous, brown, visceral and gonadal fat tissue of pregnant rats. Material and methods. In this study 24 (12 per group) Sprague Dawley rats were fed with standard (SD) or HFHSD from 3 weeks of age for 6 weeks and after mating until birth. Samples of subcutaneous and brown fat tissue were embedded in paraffin blocks and stained with haematoxylin and eosin. Data were analysed using Image J software. Results. While HSHSD did not change the surface area of adipocytes in subcutaneous and brown fat tissue, there was a significant increase in surface area in visceral and gonadal fat. Adipocytes were divided into six different classes based on their surface area. There were significant increases in the numbers of class II (800-2800 µm²) adipocytes in subcutaneous (p = 0.0008) and class I (0-800 µm²) adipocytes in brown fat tissue (p = 0.0001). In visceral and gonadal fat tissue there was no change in adipocyte classes. Discussion and conclusions. The combination of HFHSD and pregnancy increased adipocyte surface area only in visceral and gonadal fat, and resulted in adipocyte hypertrophy. The effect in subcutaneous and brown fat tissue was observed as changes in the numbers of class I and II adipocytes, and resulted in adipocyte hyperplasia. These results suggest the possibility that pregnancy induced an alternative pathway for the use of excess energy in subcutaneous and brown fat in the HFHSD group to reduce inflammation.
