mRNA Expression of Enzymes COX1, COX2, eNOS AND iNOS in Athletes Consuming Enriched Hen Eggs

Introduction: Consumption of hen eggs enriched with n-3 polyunsaturated fatty acids (n-3 PUFAs) enhances acetylcholine- dependent dilation and has positive effect on microvascular function in athletes (Kolar et al., 2021), possibly due to altered production of vasoactive metabolites. Present study aimed to examine the effects of consumption of hen eggs enriched with n-3 PUFA, lutein, selenium and vitamin E on enzymes involved in production of vasoactive metabolites. Methods: Randomized, double-blind, controlled, interventional study (Clinical Trials ID: NCT04564690) was conducted in healthy young athletes (N=14) who consumed 3 hen eggs per day for 3 weeks. Nutri4 group (N =8) consumed enriched eggs (n3- PUFAs: 1026 mg/day, lutein: 616 mg/day, selenium: 0.191 mg/day and vitamin E: 1098 mg/day), and control subjects (N=6) consumed ordinary eggs (n-3 PUFAs: 438 mg/day, lutein: 110 mg/day, selenium: 0.181 mg/day, vitamin E: 595 mg/day). Blood samples were collected before and after eggs consumption. mRNA of COX1, COX2, eNOS and iNOS enzymes expression in peripheral blood leukocyte was measured by rtPCR method (BioRAD, CFX96 Real- Time System), normalized to GAPDH housekeeping gene. ANOVA was conducted for statistical analysis with Tukey's post hoc test ; p<0.05 was considered statistically significant. Results: Three-weeks dietary intake of eggs did not cause a significant change in expression of any gene in any group. Discussion: Cyclooxygenases are the key enzymes in the conversion of arachidonic acid to prostaglandins and other lipid mediators. Prostaglandins are a group of physiologically active lipid compounds called eicosanoids that have different hormone-like effects in animals. Nitric oxide synthases are the enzymes responsible for NO generation which is an important cellular signaling molecule. It modulates vascular tone, insulin secretion, airway tone and peristalsis, and is involved in angiogenesis and neural development. Conclusion: Consumption of hen eggs did not affect gene expression of those enzymes in athletes, in baseline condition, with no exertion effort.
