Sažetak
The quest for stem cells from adult tissues is one of the major concerns of regenerative medicine research in the recent years. Hematopoietic stem cells (HSC) are the best studied and characterised adult stem cells that are used as an efficient therapy in the clinic, but they are also used as a model for development of other cell therapies for tissue regeneration. However, the hematopoietic system relies on the self-renewal, differentiation and great expansion potential of the HSC. The seminal experiments of lethally irradiated mice rescued by bone marrow transplantation are the basis of bone marrow transplantation therapy. HSC have been further characterised and isolated from the bone marrow first by Spangrude et al in 1988 and since than the isolation procedure has been improved constantly. It has been shown that HSC give rise to multipotent progenitors (MPPs) which lose self-renewal potential but can differentiate into multiple lineages while their expansion potential decreases. MPPs further give rise to oligopotent progenitors which are common lymphoid (CLP) and myeloid progenitors (MP). This process depicts the classical model of HSC differentiation as a highly regulated progression of a series of developmental cell fate evaluations. This commitment to differentiate and mature in a regulated hierarchy involves complex events mastered by several transcription factors like PU-1, Notch-1, Pax-5, and Ikaros transcription factor families. Experiments from genetically manipulated mice and in vitro studies show that they have a critical role at specific check points of the HSC differentiation. In recent years the commitment to differentiate and mature in a tree-like pyramid has been confronted by evolving single cell technologies, 3D in vitro cultures and genetically engineered mouse models showing new types of precursor cells and finding some heterogeneity among HSC.Nevertheless, how developmental regulated changes are reflected in gene expression programs is still not well understood. We are interested in the mechanisms that drive the generation of lymphocytes from HSC. We study the roles of Notch as well as transcription factors Ikaros and Aiolos in healthy human lymphocytes and in lymphocytes with abnormal development like leukemic cells. In addition to clinical studies we have also used experimental animals, mostly mice, to increase our understanding of the development of lymphocytes. Further by isolating and growing epithelial stem cells from human thymus we attempted to recreate the microenvironmental conditions that have measurable effects on lymphocyte development from HSC. Unravelling the conditions that stimulate development or cause reprogramming of HSC, opens new possibilities in research, medicine and pharmacology by understanding the mechanisms of haematopoiesis, give the possibility to get new target molecules that activate or silence single genes and processes, influence the cells that escaped a natural check point during their maturation and improve the clinical outcome of HSC transplantation.
