Sažetak
The purpose of this research was to investigate the influence of pioglitazone on neuronal loss and microglia/macrophages features in the CA2, CA3 and dentate gyrus rat hippocampal regions following traumatic brain injury (TBI) induced by the lateral fluid percussion injury. Pioglitazone was administered in a dose of 3 mg/kg at 10 min, 12, 24 and 48 hours after the brain trauma. Rats were decapitated 72 h after TBI and their brains were extracted and processed for succeeding analyses. NeuN immunohistochemistry revealed neuronal loss in the CA2 region of the traumatized rats. Digital reconstructions with skeletal analyses of the images of the Iba1 imunolabeled sections demonstrated TBI-induced changes in the microglial morphology in the hippocampal regions. Immunohistochemical analyses revealed significant increase in the number of CD68 positive cells in the CA2 and CA3 hippocampal regions in the injured rats. Pioglitazone significantly diminished neuronal loss in the CA2 region, but it did not affect TBI-induced changes in the microglial morphology or the number of CD68 cells. Results of this study show new favorable pioglitazone effect in the model of TBI in the rat.
