Hedgehog-GLI signaling pathway in 3D model of prostate cancer

Prostate cancer is the second most frequent cancer diagnosed in men worldwide. Early stages of the disease can be effectively treated while still localized but management of advanced stages is limited due to the development of biological mechanisms of resistance to medication. Androgen receptor (AR) signaling is the main pathway responsible for prostate cancer cell growth. Therefore, androgen deprivation therapy (ADT) has been the standard treatment option for patients with advanced forms of prostate cancer. However, for most of the patients ADT therapy is effective only at the beginning, after which there is a significant progression to a poor-prognosis stage called castration-resistant prostate cancer (CRPC). In addition, the role of Hedgehog-GLI (HH-GLI) signaling pathway, as its aberrant activation in adult cells has been linked with the development of prostate cancer, must be considered as well. Therefore, the aim of this research was to better understand the differences in HH-GLI signaling pathway in two-dimensional (adherent) and three-dimensional (spheroid) models of prostate cancer.
