Fluorescent Self-assembled Nanostructures from Pyrene-derived Retropeptides: Fluorescence Properties and Application in Cell Imaging
Self-assembled peptide nanostructures are very promising biomaterial with huge potential in several biomedical applications. Fluorescence imaging is emerging as promising and powerful means for monitoring various bioactive molecules in living systems, which is thanks to its remarkable advantages, such as easy operation, high sensitivity and high temporal-spatial resolution. In aim of design of fluorescent self-assembled nanostructures, a novel series of mono- (9a-c) and bis-1, 2, 3-triazolepyrene-appended (4a-c) retropeptides were designed and prepared via the standard copper-catalysed azide-alkyne cycloaddition (CuAAC). Their photophysical properties and morphology were fully investigated by UV–vis and fluorescence spectroscopy, and AFM. Bis-1, 2, 3-triazole derivatives were prone to self- assemble into fluorescent nanoparticles in aqueous media, which was further confirmed by fluorescence spectroscopy, AFM and confocal microscopy. Most of the bis-1, 2, 3-triazolpyrene signals in cells are grouped in clusters. To determine a possible cytotoxic effect of the novel 1, 2, 3-triazoles on human cell lines, the MTT assay was performed. While mono-1, 2, 3-triazoles did not show significant cytotoxicity, bis-1, 2, 3- triazoles showed reduced cell proliferation in the concentration range of 10-5 - 10-7 M.
