Electrochemical detection of polyhenols in food samples

Polyphenols are the significant plant compounds with antioxidant activity and are the subject of increasing scientific interest because of their beneficial effects on human health. They have shown anticarcinogenic, antimutagenic, anti- inflammatory and antiallergic properties. The most important polyphenols are the ones which come from routine consummation of fruits and vegetables so it is important to detect concentrations of polyphenols in real samples. Protocatechuic acid and p-hydroxybenzoic acid are natural polyphenols mostly found in green tea, acai oil, plums, grapes, coconut, wine etc. The main goal of this study was to detect protocatechuic acid and p- hydroxybenzoic acid in model systems and real samples with differential pulse voltammetry. The experiments were performed in a three electrode voltammetric cell with glassy carbon as working electrode, Ag/AgCl as reference electrode and platinum wire as counter electrode. Before each measurement, the system was purged with high purity argon Ar5 (fi (Ar) = 99.999 %). Differential pulse voltammograms have shown one oxidation peak of investigated polyphenols in pH range (pH = 3-8) which correspond to oxidation of hydroxyl group(s). The highest peak current was around pH = 4 (protocatechuic acid) and around pH = 6 (p-hydroxybenzoic acid) while it decreased in more acidic and alkaline medium. It was also determined that the anodic peak current of investigated polyphenols increased with the increase of their concentration (linear response was obtained in the concentration range from 1 uM to 600 uM). Both polyphenols were detected in real samples where their quantity varied from 2 uM till 9 uM.
