Sažetak
Endocrine disruptors (EDCs) are defined as an exogenous chemical or mixture of chemicals that can interfere with any aspect of the hormone action. Scientific Statement of The Endocrine Society presented evidence of EDCs effects on male and female reproduction, breast development and cancer, prostate cancer, thyroid function and more. In this research, we have investigated effects of several commonly used pharmaceuticals that are suspected to have endocrine disrupting properties (paracetamol, ibuprofen, fluoxetine and carbamazepine) and polystyrene micro- and nanoparticles (PSMPs and PSNPs) on estrogen receptor activity using T47D-KBluc cell line. This cell line is stably transfected with a triplet ERE (estrogen-responsive elements)-promoter-luciferase reporter gene construct and therefore can be used to screen chemicals for estrogenic and anti-estrogenic effects. After thorough cell viability tests (MTS assay and Annexin V/Propidium Iodide Apoptosis Assay), three concentrations of each compound and two concentrations of PSMPs/PSNPs were chosen for further experiments. Preliminary results have shown that the tested chemicals and nanoparticles show both anti-estrogenic and estrogenic effects and that these effects are modulated when drugs are mixed with nanoparticles. Mixtures with estrogenic effects could lead to development of ER positive breast cancers (about 80% of breast cancers are ER positive). This research will provide important new insights on impact and safety of pharmaceuticals and polystyrene nanoparticle mixtures, seeing that these mixtures can be found in environment.
