Ferrocene Chromophore as a Circular Dichroism Probe for the Helicity Sensing of Peptides
Circular dichroism (CD) probes are achiral molecules that undergo chiral perturbation in an asymmetric environment and convert the chiral information into a strong and characteristic CD signal. To reach guidelines for a design of ferrocene-based probes eight stereoisomers of monosubstituted ferrocene, Boc-Pro-Pro-Ala-NHFc, have been prepared and conformationally characterized in depth. A combined strategy utilizing experimental (IR, NMR, CD) and computational (DFT) techniques is illustrated on tripeptides to investigate their chiroptical properties. The origin of the sign in CD spectra near the absorption maximum of a ferrocene chromophore is strongly correlated with the sign of the dihedral angle describing deviation of the directly attached amide plane from planarity of the cyclopentadienyl ring. The results of this and previous studies should be considered as an important step towards the use of ferrocene-based CD probes for the assignment of the screw-sense preference of short peptides.
