DNA barcoding reveal larval fish assemblages in the eastern Adriatic Sea (eastern Mediterranean)

During the scientific survey (8-29 July 2020), ichthyoplankton samples were collected with a WP2 sampler at sixty sampling stations along the eastern Adriatic. Ichthyoplankton samples were put in a marked plastic container and preserved in 96% absolute ethanol. The presence of fish larvae was noted in 39 stations. Each collected larva was measured and photographed under the stereomicroscope. Afterwards, gDNA from individual larvae was isolated using standard proteinase K digestion followed by phenol:chloroform extraction protocol. The partial fragment of cytochrome oxidase subunit I (COI) gene was amplified by PCR using primer pair FishF1/FishF2. The obtained sequences were analysed for similarity with other known vertebrate sequences using Blast Local Alignment Search Tool. A total of 194 larvae belonged to 14 larval fish taxa. Overall, anchovy larvae were the dominant ones (75%). Application of molecular taxonomy reveals us, for the first time, the occurrence of new Gobiidae species in the Adriatic.
