Sažetak
Introduction: Yeast Saccharomyces cerevisiae is an ideal model organism for unrevealing molecular mechanisms of the stress response provoked by metals. In this research to study the action of metal ions released from orthodontic materials (nickel-titanium archwires and steel brackets) Saccharomyces cerevisiae yeast, strain W303 was used. Material and Methods: YPD media were used for the preparation of metal-ions enriched media (1 cm of archwire - 1 mL of medium ; on 20 brackets 28 mL of media) simulating oral cavity conditions (continuous stirring at 40 rpm, 37 °C, 3 days). Then the yeast was added and grown to an early stationary phase. Cell viability was determined by counting the yeast cell number in a Bürker- Türk chamber and using a Luna-FLTM cell counter, while for metabolic activity determination Live- DeadTM yeast Viability kit was used. Mitochondrial activity was measured as oxygen consumption rate using the Agilent Seahorse XF Cell Mito StressTM test. Results: A reduced number of cells grown on metal-ions enriched media was observed, with the largest number reduction in the cells grown on archwires + brackets – ions enriched medium. In addition to the decrease in the cell number, these cells were smaller in diameter with a higher tendency to grow in clusters. Comparing the metabolic activity by the metal sequestration ability, and by active vacuoles, there are no differences in the metal- ions treated cells compared to the control. Cells grown on metal- ions enriched media have altered all cellular respiration parameters (basal respiration, spare respiratory capacity, proton leakage) measured as the oxygen consumption rate of the cells. Conclusion: Altered mitochondrial activity of metal-ions treated cells indicates that yeast mitochondria were the most susceptible to the harmful action of metal ions, while the activity of their vacuoles was preserved.
