Kinetic analysis of purple non-sulphur bacteria Rhodovulum adriaticum DSM 2781 growth on carbon sources from lignocellulosic hydrolysates

Purple non-sulfur bacteria isolated from different habitats are interesting in sustainable processes for production of hydrogen, in medicine for synthesis of biochemicals such as aminolevulinic acid, for production of eco-friendly biopolymers (PHB) and biomass along with synthesis of specific compounds such as carotenoids and pigments. As they can grow phototrophic and photoheterotrophic, various cultivation have been recognized for years. However, in order to achieve an ecologically and economically sustainable production, it is necessary to select the appropriate feedstock and cultivation conditions. The aim of this study was to determine growth kinetics, substrate utilization and the ability of Rhodovulum adriaticum DSM 2781 to grow under different carbon sources obtained after acidic pre-treatment of waste lignocellulosic feedstock in high-pressure reactor. Selected strain can be cultivated in broad range of glucose and/or xylose concentrations, and highest biomass yields were achieved with glucose as a carbon source, although growth is partially inhibited when carbon source concentrations are above 10 g/L. Considering certain limitations under photosynthetic conditions, the chemoheterotrophic dark/aerobic growth on glucose and xylose from pre-treated lignocellulosic biomass as carbon sources and with DAP and yeast extract as nitrogen sources in a stirred tank bioreactor was also investigated. During batch cultivation highest bioprocess productivity of 0, 154 g L-1 h-1 was achieved with of biomass yield of 3, 7 g L-1, YX/S = 0.521 g g-1 and with observed growth rate (µ) of 0.551 h-1.
