Oroxilum indicum has an extensive history in the Indian traditional medicine system. The root and stem bark of this plant is well accepted for the treatment, prevention, and cure of various types of diseases. The bark of the plant has a high content of flavonoids and other phenolics, which are known to be responsible for exhibiting various therapeutic effects. The role of O. indicum in liver disease is well known, but until now, the mechanism has not been well studied. In the present study, the stem bark of O. indicum was extracted with ethanol using a hot extraction method and their phyto-chemical analysis was performed. Further, the plant extract was also used to evaluate their role in reducing oxidative stress and β-glucuronidase enzyme activity. The antioxidant study showed that extract has the capability to down-regulate oxidative stress, while β-glucuronidase assay displayed IC50 at 174.36 μg/mL. Additionally, we selected the major polyphenolic phyto- compounds derived from the bark of O. indicum for in silico ADME and docking studies. The ADME study reflected the optimal pharmacokinetic profile of the compounds. However, the docking experiments confirmed that all the selected phyto-compounds have tendency to interact with β-glucuronidase receptor.
