Immunohistochemical study: could SOX17 gene be new biomarker for testicular seminoma?
The most frequent testicular cancer is a testicular germ cell tumour (TGCTs). Despite the fact that testicular neoplasia represents only 1% of all neoplasia, TGCTs affect young male population thereby representing a well- recognized challenge in the frame of male reproductive health. TGCTs are classified into seminoma and nonseminoma. In the routine diagnosis of TGCTs specific biomarkers as OCT ¾ are used. OCT ¾ is a sensitive and specific marker for seminoma and embryonal carcinoma. Nonetheless, investigating other genes and their specific expression in seminoma could improve patient management. In our research, we compared the expression of OCT ¾ and SOX17 in seminoma tissue to investigate its potential as biomarker for testicular seminoma. In our study, 52 archive seminoma paraffin-embedded tissue was collected, cut on the 5µm and immunohistochemically stained. To visualize SOX17 and OCT¾, specific antibodies were used. Morphometric analysis for OCT ¾ and SOX17 expression was performed by pathologists. Both genes, SOX17 as well as OCT ¾ were expressed in seminoma tissue. However, out of 52 tissue samples, in more than 50% of seminoma cells, SOX17 present in just 11 patients while OCT ¾ in 18 samples analysed. From the findings it can be concluded that SOX17 could be a potential marker for testicular seminoma and used aside with already known biomarkers. This presents a potential to improve diagnosis of seminoma and seminoma patient management overall.
