Determination of cannabinoid content in industrial hemp

Hemp (Cannabis sativa L.) is one of the oldest plants which is widely used in different industries. It contains cannabinoids some of which are psychoactive like THC. In the Republic of Croatia is allowed to breed varieties of industrial hemp in which the THC content does not exceed 0.2 %. This paper presents the results of determining the THC content in different varieties of industrial hemp during 2020, in order to determine health safety. In addition to THC the following cannabinoids were analyzed in the samples: CBD, CBN, and CBG. From the homogenized sample the cannabinoids were extracted with absolute ethanol. They were analyzed by gas chromatography with a flame ionization detector (GC-FID) and quantified by the method of internal standard (TBA - tribenzylamine) using a calibration curve. The method was developed in the laboratory, validated and accredited. RSD for repeatability of sample injection is from 0.30 to 0.62 %, while repeatability of sample preparation is ranges from 0.88 to 2.17 %. In laboratory during 2020 were analyzed 103 samples of hemps, of which 10 samples showed that the mass fraction of THC was higher than 0.2 %. Samples with the content of THC higher than 0.2 % were analyzed by confirmatory technique, liquid chromatography coupled to mass spectrometry LC-MS / MS. This technique confirmed that of all the positive samples on GC-FID, only in one sample content of THC was higher than 0.2 %.
