Quantitative changes in proteomes of Listeria monocytogenes and Yersinia enterocolitica treated with quaternary ammonium salts based disinfectants

Pathogenic, gram-positive bacteria, Listeria monocytogenes as well as the gram-negative Yersinia enterocolitica are often involved in food spoilage and food-borne infections. Therefore, they are frequently used as a model microorganism for analyzing food contamination. Antimicrobial agents based on quaternary ammonium salts (QAC) are very effective and widely used class of disinfectants. Their extensive use has, however, resulted in increased antibacterial resistance. In this work, label-free quantitative gel-free proteomic investigations by LC-ESI MS/MS were used to investigate the influence of four different QAC based disinfectants on L. monocytogenes and Y. enterocolitica. MaxLFQ, the label-free quantification (LFQ) module of MaxQuant software was employed and the computational analysis of data was performed with Perseus based on LFQ values of proteins. Up- and down-regulated proteins after the growth under the inhibition were identified using Uniprot database and their cellular localizations and functions were determined by gene ontology searching using web- based tool QuickGo. Proteome changes presented demonstrate different mechanism of action of these disinfectants against gram-positive and gram- negative bacteria and contribute to the investigation of the virulence and pathogenicity of food borne bacteria and their survival under stress conditions.
