Sažetak
Gastrulating rat embryo-derived teratoma in vitro system has been used for studies of developmental toxicity and anti-tumor agents 1, 2, 3, 4. Our aim was to associate the impact of histone deacetylase epigenetic drug valproate on histone acetylation and apoptosis in teratomas with biomarkers detected in spent media metabolomes by the infrared spectroscopy. 9.5-days-old Fisher rat embryos were cultivated for 14 days in Eagle's MEM and 50% rat serum at the air-liquid interface with or without 2mM valproate. In developed teratomas histone acetylation was determined by western blotting and apoptosis by immunohistochemistry using a cleaved caspase-3 monoclonal antibody and stereological quantification (volume density (Vv)) of the positive signal. Spent media metabolomes were analyzed by the Fourier Transform Infrared (FTIR) spectroscopy and multivariate statistical analysis. Valproate significanty increased histone acetylation and Vv of the apoptosis marker. PCA scatter plot of FTIR results separated all spent media accordingly to treatments and lenghts of culture period. An elevated level of acetylation FTIR biomarkers, the amide I α-helices and the elevated ratio of A(CH3)/A(CH2 together with biomarkers of apoptosis, a higher intensity of the 1625 cm-1 absorption band for the parallel β-strand structure and CH2 vibrations of lipids at 2853 cm-1 were detected. We may conclude that FTIR spectroscopic analysis of the spent media metabolomes reflected the processes occuring in the embryo treated with the antiepileptic/teratogen/anti-tumor drug valproate and may upgrade this biological system for faster screening for embryotoxic/anti-tumor agents.
