Chronic stress causes age specific changes of brain lipid rafts in Sprague Dawley female rats
Methods: Young (6-months-old) and old (14.5- months-old), male and female Sprague-Dawley rats were submitted to three sessions of 10-day CS protocol with recovery time between and at the end of the last session. Brain sections were used for triple-staining (Np/GD1a/CTB) and analyzed in the hippocampal areas CA1 and dentate gyrus (DG) by confocal microscopy and 3D tissue reconstruction. Statistical differences were determined with Statistica 12 software. Kruskal-Wallis test with post hoc analysis by Mann-Whitney U test were applied. Results: In CA1, the colocalization (Np/GD1a/CTB) was lower in all CS groups compared to control groups, except for the old females. Significance was reached in young CS males (p=0.002) and females (p=0.004) compared to control groups. Young CS females had increased colocalization compared to young CS males (p=0.004). In DG, the colocalization was lower in all animal groups upon CS, significantly in old males compared to control group (p=0.005). 3D reconstruction of CA1 and DG revealed less diffused and decreased expression of Np and CTB upon CS in old animals. Conclusions: Changes in lipid rafts content upon CS, especially decrease in Np/GD1a/GM1 colocalization are age and sex specific. Lipid rafts destabilization by CS could cause synaptic instability through changed interactions with Np.
