A comparison of activating agens for efficiency and stability of aldolase immobilized on magnetic nanoparticles
Enzyme immobilization is a promising technique for improving enzyme stability while retaining its activity. It often includes binding of an enzyme to a solid carrier, by employing covalent bonds or by adsorption. If using covalent bond, the carriers should be activated with appropriate activating agents that act as a link between carrier and enzyme. The aim of this work was to immobilize aldolase on silica coated magnetic nanoparticles. For that purpose carrier was synthesized using solvothermal method and three different types of activating agents, glutaraldehyde, benzoquinone and 3-Glycidyloxypropyl trimethoxysilane (GPTMS), were used (Figure 1). The features of immobilized aldolase were examined in the reaction of double aldol addition for production of statin side chain precursors, a valuable pharmaceutical intermediate. Retained activity and enzyme stability of immobilized aldolase was compared with that of a free enzyme.
