Synthesis and Evaluation of a QS-17/18-Based Vaccine Adjuvant

We have synthesized QS-17/18 analog 7 and evaluated its adjuvant activity in the formulation with rHagB antigen. Compound 7 and QS-21 analogs 5 and 6 are presumably the major components of the widely studied semi-synthetic saponin adjuvant mixture GPI-0100. Based on its IgG subclass antibody response assessments, adjuvant 7 shows an adjuvant activity profile similar to that of GPI-0100 which potentiates a mixed Th1- and Th2-like response to rHagB. Their profiles are different from those of QS21 analogs 5 and 6 that probably only induce a Th2-like immunity. Combination of QS-21 and QS- 17/18 analogs does not show a synergistical effect. These results suggest that compound 7 might be the main active component of GPI-0100 and is responsible for the favorable adjuvant property of GPI-0100.
