Application of ICY software to motility analysis in cadmium polluted algal cell cultures

Heavy metals are widely dispersed water pollutants due to industrial, agricultural and urban development and they have a significant impact on organisms in water ecosystems. Microalgae cells are mostly very sensitive to changes in the environmental factors such as temperature, salinity, light intensity and also the presence of toxic pollutants in their environment, which are responsible for their growth and physiology, and also autonomous cell features such as photosynthesis and motility. The aim of this work was to examine the algal cell motility in stress conditions. As model organisms we used three species of microalgae Dunaliella tertiolecta (Chlorophyceae), Rhodomonas maculata (Cryptophyceae) and Tetraselmis suecica (Chlorophyceae), that differ in the structure of the flagellar apparatus. Microalgae in growth medium were exposed to cadmium concentration of 1000 µg/L. The mathematical model enabled to determine free cadmium concentration that act as toxicants to algal cell [1]. We employed open- access software Icy (http://icy.bioimageanalysis.org/) to perform motility analysis of cells in the growth medium. The results will be presented in terms of cell velocity and search path of three microalgal species. This study could contribute to better understand cell behavior change when exposed to stress without any sample pretreatment.
