Is Na/K-ATPase expression in pig’s small intestine gender dependent?
Na/K-ATPase is an enzyme found in all animal and human cells, which confirms its essential role in the physiology of cells such as the regulation of intracellular homeostasis of sodium and potassium ions, membrane potential, pH and cellular absorption of monosaccharides and amino acids in various tissues. It is mainly located on the basolateral cell membrane of the enterocytes, which facilitates the maintenance of the sodium ion gradient required for transepithelial transport of nutrients with sodium dependent cotransporters located on the brush border membrane of enterocytes. There are numerous studies on the Na/K-ATPase expression in pigs, particularly in kidneys, while the data on the expression in the small intestine are rare. The aim of this study was to determine the influence of sex hormones on the Na/K-ATPase expression in the small intestine of pigs. The study was conducted on 16 pigs Swedish Landrace breed divided into 4 groups (males, females, castrated males and ovariectomized females). Tissue samples were collected from the duodenum, jejunum and ileum and were analyzed for the Na/K-ATPase expression by Western blotting of isolated total cell membranes and by immunostaining of tissue cryosections using specific monoclonal antibody. Preliminary results of both used methods indicate the existence of gender dependent differences in the Na/K-ATPase expression in pig’s small intestine. Differences in the expression in pigs of the same gender but different groups (male/castrated male and female/ovariectomized female) suggest that sex hormones affect the Na/K-ATPase expression in the small intestine.
