Azithromycin-loaded liposomes for the treatment of sexually-transmitted infections: anti- chlamydial and biocompatibility studies

This study aimed to determine anti-chlamydial activity of the several types of azithromycin (AZI)-loaded liposomes as well as their compatibility with cervical HeLa cells. Propylene glycol AZI-liposomes (PGLs) and deformable propylene glycol AZI-liposomes (DPGLs) were prepared by polyol dilution method, while conventional AZI-liposomes (CLs) were prepared by proliposome method [1]. Following physical characterization at mimicked cervical conditions, all the liposomes were tested against intracellular C. trachomatis. The infected HeLa cells were treated with the different AZI-liposomes or AZI-solution (control), and the anti-chlamydial effect was assessed by determining minimum inhibitory concentrations (MICs) using direct quantitative polymerase chain reactions (qPCR) measurements [2]. The in vitro biocompatibilities of AZI- liposomes were evaluated by MTT test following incubation of the liposomes with the HeLa cells during 24 hours. All AZI-liposomes were negatively surface charged and in the size range 195-270 nm. PGLs and particularly DPGLs were characterized with elastic (deformable) bilayers, while bilayers of CLs were quite rigid due to the presence of hydrogenated phospholipids. In vitro antibacterial studies showed that DPGLs were the most potent against intracellular C. trachomatis with MIC value of 0.0312 µg AZI/ml, followed by PGLs (0.0625 µg AZI/ml) and CLs (0.125 µg AZI/ml). The performed biocompatibility studies proved that all the AZI-liposomes were biocompatible with the HeLa cells. Even at the highest tested AZI concentration (39.5 µg/ml), the viability of the cells remained above 70%. The performed studies indicate the influence of bilayer elasticity/rigidity of AZI-liposomes on tolerability with the cervical cells and anti- chlamydial activity.
